17-AAG, HSP90 inhibitor

So I have a bit of a theory here. I have not heard anything like this talked about on hair loss sites, but this seems like a reasonable idea.

That article shows that in prostate cancer even with anti-androgens in place, tumors will eventually relapse and continue to grow. This is a result of upregulation of ARs + mutations that eventually lead to an AR that fires without any androgens even present. They tracked this down to something called HER2 which creates several pathways that eventually create loops that fire the AR without androgen.

I have a strong feeling that this exact same mechanism is why propecia loses effectiveness after 5 years or so of use.

I read this this morning, but if I remember correctly, HSP90 is a cofactor that is responsible for both the production and destruction of various proteins, one of which is the AR. 17-AAG is medicine that forces the HSP90 into destructive mode, which actually REMOVES several pathways (including the AR itself!) from the cell.

Not only do I think this could result in extended propecia effectiveness, but maybe it could even work by itself? Anybody with more experience and/or knowledge please chime in.

The second article seems indicates a similar effect from resveratrol. I have a question about that though, based on wikipedia, the most effective way of administering resveratrol (without injections) is orally (but NOT swallowing). Is this safe? I wonder what my pills taste like.

Talks about how Resveratrol + 17-AAG = more effective 17-AAG due to Resveratrol controlling HSP70 which is upregulated while you are on 17-AAG...

Not entirely sure what all that means.

It is still in trials but apparently it is the "protector" of LOTS of proteins in the cell, so getting rid of it (HSP90) isn't such a good call. Additionally, the drug seems to have a higher affinity for a mutated form of HSP90 found in cancer cells.

I still think there is potential in this with a less potent and low dosage approach. I'll try and find some natural HSP90 inhibitors.

Is a patent to STOP hair growth, which is a bit odd, but it mentions the molecule 17-AAG was derived from and Resveratrol in the same sentence as potentials for some step in the patent.

Based on that and the article showing Resveratrol shows similar effects as 17-AAG, I'm guessing that it works in a similar fashion. SOOO I'm going to have to get back on some res.

so i broke a resveratrol pill and tried to let it absorb in my mouth.... #$%! wretched. it tasted like charcoal.

maybe i'll put it in some water and swish that around my mouth for a minute or two
