Many therapeutic modalities has been developed to improve and enhance the hair growth. Among many chemical and natural compounds, some potentials were observed from certain synthetic peptides such as tripeptide-copper complex consist of glycyl-histidyl-lysine-Cu2+ in which both wound healing and in vitro hair growth were notable. In present study the hair growth stimulatory effect of peptide-antioxidant-mineral compounds were investigated and screened. Seven peptide complex derivatives (VitC-AHK, VitC-AHK-Cu2+, VitC-DAHK-Cu2+, VitC-KGHK-Cu2+, DAHK-Cu2+, AcDAHK-Cu2+, KGHK-Cu2+) were synthesized in which either tri- or tetrapetide linked to both side with antioxidant vitamin C and copper ion. Among them a copper ascorbyl phosphate succinyl tripeptide-34 (VitC-AHK-Cu2+; named as Alotide) showed favorable effect on dermal papilla cell proliferation with no cytotoxicity. This peptide showed remarkable hair re-growth pattern in C57BL/6 mouse than others following topical application. NMR structure analysis suggest that this peptide maintains stabilized molecular bond with copper ion. Thus hair growth promoting effect might be related to structure-activity of copper-tripeptide.
